Discovery history

Developed as a soluble activin type IIB receptor fused to an IgG1 Fc domain to bind myostatin and related ligands and increase skeletal-muscle mass.

Proposed Mechanisms

03

Reported / Studied Uses

04

Human Evidence

Early trials showed increased lean mass and muscle volume. A Duchenne muscular dystrophy trial was stopped early because of epistaxis and telangiectasia, and functional benefit was not established.

05

Animal Evidence

Animal studies show substantial increases in skeletal-muscle mass through blockade of myostatin and related ligands.

Species translation to humans is uncertain.
06

Safety & Unknowns

development-discontinuedvascular-adverse-effectsepistaxistelangiectasiaunapproved-product-risk
Clinical development identified nosebleeds and telangiectasia consistent with broad activin-family ligand trapping. Long-term vascular, reproductive and systemic safety was not established.
